Introduction
A series of 6-amino-5-cyano-4-phenylpyrano[2,3-c] pyrazole-4-carboxylic acid esters was synthesized via the four-component one-pot reaction of ethyl acetoacetate, hydrazine, methyl phenylglyoxylate, and malononitrile in the presence of InCl3 in 50% EtOH medium under ultrasound irradiation at 40 °C for 20 min. The ever-growing need for new chemicals is being met by organic chemists’ efforts to synthesize novel structures bearing various rings such as pyrans, pyrazoles, and pyrimidines [1]. The pyranopyrazole ring, a productive source of biologically active molecules [2], is attracting increasing interest from chemists. Heterocyclic compounds containing pyranones and pyrazolines have been widely investigated for their potentially useful bioactivity [3, 4]. The synthesis of pyrano[2,3-c]pyrazoles has received much attention owing to their antimicrobial [5], anti-inflammatory [6], anti-tumor, and antifungal [7] activities. They have also been useful in combating schizophrenia, Alzheimer’s, and mycolonous diseases [8]. Additionally, pyranopyrazole derivatives have been reported to show vast utility in biodegradable agrochemicals, pigments, cosmetics, and photoactive materials.[9]. Much recent research has focused on the synthesis of multi-substituted dihydro-pyrano[2,3-c]pyrazoles with various functional groups [10].

Multicomponent reactions (MCRs) are widely employed to make bioactive scaffolds [11] and complex heterocycles [12]. They involve three or more reactants that form three or more chemical bonds incorporated into a single molecule via one-pot domino or tandem reactions. Many studies have proved that MCRs show atom economy, short reaction times, and high efficiency. They are thus widely applied in synthetic chemistry, medicine, and agrochemistry [13].
However, establishing a convenient and efficient approach for the synthesis of pyrano[2,3-c]pyrazoles remains challenging. Various methods have been reported for the synthesis of substituted pyrano[2,3-c]pyrazoles involving different metal catalysts such as nano-TiO2/H14[NaP5W30O110] [14], P4VPyCuI [15], and ZrO2 NPs [16]. In addition, multi-substituted pyrano[2,3-c]pyrazole derivatives have been obtained in good yields under catalysis by triethylamine [17] and metalfree or solvent-free conditions [18, 19]. Although most of the above-mentioned methods offer some advantages, they also suffer from disadvantages such as long reaction times, operational complexity, and the use of rare or expensive catalysts. InCl3 is an effective Lewis acid catalyst [20, 21] and has a high affinity toward acetylenic
bonds [22], intermolecular [23] and intramolecular [24] reactions of arenes with allyl acetates, and allyl bromides.
Ultrasound irradiation is increasingly being used in synthetic chemistry because it allows quicker and more selective reactions with excellent yields in comparison with conventional methods [25]: Ultrasonic-accelerated reactions complete more quickly and under more moderate conditions [26]. Therefore, we designed a facile and efficient method for the synthesis of pyrano[2,3-c]pyrazole derivatives via MCRs in 50% EtOH catalyzed by InCl3 under ultrasound irradiation (Scheme 1). The products contained various functional groups that have various useful applications.

Results and discussion
To establish optimal reaction conditions, the four-component one-pot reaction was explored using ethyl acetoacetate 1a, hydrazine hydrate 2, methyl phenylglyoxylate 3a, and malononitrile 4 in 50% EtOH catalyzed by InCl3 under ultrasound irradiation as a model reaction. This reaction did not occur in the absence of InCl3 (Table 1, entry 1). Different concentrations of catalyst (5, 10, 20, and 30 mol%) were then screened for this reaction, and the product 5a was obtained in excellent yields (85–95%, Table 1, entries 2–5). The optimal concentration of 20 mol% InCl3 obtained the highest yield with a relatively small amount of catalyst (entry 4). Various other catalysts (Et3N, piperidine, TEBA, Na2CO3, and KOH) were then tested with the model reaction (Table 1, entries 6–10), showing that organic alkali produced higher yields than inorganic bases. Overall, catalyst screening indicated that the best yield (95%) was obtained using InCl3 (20 mol%) (Table 1, entry 4).
Six different solvents were then screened to find the best for the synthesis of compound 5a using InCl3 (20 mol%) catalyst under ultrasound irradiation at 40 °C for 20 min. The results (Table 2, entries 1–6) show that 50% EtOH was more effective than 99% EtOH, H2O, THF, CH2Cl2, and MeOH (Table 2, entry 2, 95% yield). Furthermore, the model reaction with InCl3 (20 mol%) under ultrasound irradiation at 40 °C was performed for 5, 10, and 30 min (entries 7–9). Changing the reaction time did not improve the yield originally obtained after 20 min (Table 2, entry 2). The reaction temperature was next varied (from 20 to 60 °C, Table 2, entries 10–12) under ultrasound irradiation, showing that the yield was improved as the temperature increased to 40 °C, above which it reached a plateau.

Therefore, the most suitable reaction temperature is 40 °C. Finally, the scope of this process was tested by synthesizing multi-substituted pyrano[2,3-c]pyrazole-4-carboxylic acid ester derivatives in the presence of InCl3 (20 mol%) in 50% EtOH under ultrasound irradiation at 40 °C for 20 min.
The results summarized in Table 3 show that compounds 5a–5t were successfully obtained under the optimized reaction conditions. Whether electron-rich or electron-deficient hydrazine derivatives were used, they were well tolerated in this protocol to produce the corresponding compounds in excellent yields (80%–95%). Ultrasonic irradiation played an important role in shortening the reaction time and enhancing the yields. The structures of all synthesized compounds 5a–5t were corroborated by NMR, IR, and ESI–MS spectroscopy. Detailed spectral data and spectra of all compounds are listed in supporting information. Scheme 2 depicts the most plausible reaction mechanism for the formation of product 5. One molecule of malononitrile is first condensed with the methyl benzoylacetate, catalyzed
by InCl3, to afford intermediate C. It is reasonable to assume that A results from the initial reaction of hydrazine 2 with β-ketoester 1. Pyrazolone A loses a proton under InCl3 to give B. Subsequent nucleophilic attack by the active methylene of B on C gives intermediate D, which further produces the desired product 5 followed by tautomerization.
Conclusion
In summary, a series of 6-amino-5-cyano-4-phenylpyrano[2,3-c] pyrazole-4-carboxylic acid esters was synthesized via the four-component one-pot reaction of ethyl acetoacetate, hydrazine, methyl phenylglyoxylate, and malononitrile in the presence of InCl3 (20 mol%) in 50% EtOH medium under ultrasound irradiation at 40 °C for 20 min. Promising characteristics of the presented methodology are efficiency, simplicity, high yield, short reaction time, ease of product isolation, and agreement with green chemistry protocols, making it a useful and attractive process for the synthesis of pyrano[2,3-c] pyrazole derivatives.
Experimental
Reagents and apparatus
Melting points were determined on a Buchi B-540 digital melting point apparatus using an open-ended capillary tube and are uncorrected. A Bruker (Advance) 400 MHz NMR instrument recorded 1H and 13C NMR spectra at 400 MHz and 100 MHz using TMS as an internal standard and DMSOd6 as solvent. Chemical shifts are given in parts per million, and the coupling constants are given in hertz. Infrared (IR) spectra were recorded with a Bruker Equinox55 instrument using KBr pellets. Band positions are reported in reciprocal centimeter (cm−1); HRMS was conducted with a Micromass Q-TOF Global Tandem
Mass Spectrometer, and MS spectra were detected with an MSD1–MS instrument (LC–MS, ESI source). Sonication was performed in a Branson CQX ultrasonic cleaner (frequency 25 kHz; nominal power 250 W). The chemicals used in this study were of analytical purity and were purchased from TCI, Alfa, Acros, etc. General procedure for the four-component one-pot synthesis of 6-amino-5-cyano-3-methyl-4-phenyl-2,4-dihydro-pyrano [2,3-c]pyrazole-4-carboxylic acid methyl ester 5a–5t To a 25-mL Erlenmeyer flask charged with a mixture of ethyl acetoacetate (2.0 mmol) and hydrazine hydrate (2.0 mmol) was added dropwise a solution of methyl benzoylformate (2.0 mmol), malononitrile (2.0 mmol), and InCl3 (20 mol%) in 50% EtOH (5 mL). The reaction flask was then irradiated in the water bath of an ultrasonic cleaner (frequency 25 kHz;nominal power 250 W) at 40 °C for 20 min). The reaction was monitored by TLC. After completion of the reaction, the mixture was allowed to cool to room temperature, and an
equal volume of water was added. White or colorless crystals were obtained through filtration via sequential washing with water and ethylacetate. A pure product was obtained by crystallization in 95% EtOH.
